Production of isoamylase by Escherichia intermedia.

چکیده
A culture of a coliform bacteria isolated from soil produced isoamylase when grown in peptone-maltose-salts broth. The enzyme preparation was partially purified. Its similarity with isoamylase produced by Aerobacter aerogenes was established by studying its action upon various polysaccharides. A study of the organism producing isoamylase showed that it differed from A. aerogenes in some of its physiological properties. This organism was identified as Escherichia intermedia from morphological and cultural characteristics.

Doubling Power Output of Starch Biobattery Treated by the Most Thermostable Isoamylase from an Archaeon Sulfolobus tokodaii
Biobattery, a kind of enzymatic fuel cells, can convert organic compounds (e.g., glucose, starch) to electricity in a closed system without moving parts. Inspired by natural starch metabolism catalyzed by starch phosphorylase, isoamylase is essential to debranch alpha-1,6-glycosidic bonds of starch, yielding linear amylodextrin - the best fuel for sugar-powered biobattery. However, there is no ...
متن کاملExtra-cellular Isoamylase Production by Rhizopus oryzae in Solid-State Fermentation of Agro Wastes
Extra-cellular isoamylase was produced by Rhizopus oryzae PR7 in solid-state fermentations of various agro wastes, among which millet, oat, tapioca, and arum (Colocasia esculenta) showed promising results. The highest amount of enzyme production was obtained after 72 h of growth at 28°C. The optimum pH for enzyme production was 8.0. Among the various additives tested, enzyme production increase...
متن کاملRole of the Escherichia coli glgX gene in glycogen metabolism.
A role for the Escherichia coli glgX gene in bacterial glycogen synthesis and/or degradation has been inferred from the sequence homology between the glgX gene and the genes encoding isoamylase-type debranching enzymes; however, experimental evidence or definition of the role of the gene has been lacking. Construction of E. coli strains with defined deletions in the glgX gene is reported here. ...
متن کاملProduction of Isoamylase from Rhizopus oryzae in Submerged Fermentation
This work mainly focus on factors relevant for improvement of enzyme hydrolysis by using Rhizopus oryzae MTCC 9642 in submerged fermentation. Several cultural conditions were examined to access their effect in optimizing enzyme production. The Rhizopus oryzae PR7 MTCC 9642 showed best growth at 28°C within a broad pH range of 6.0 to 10.0 with an optimal temperature and pH of isoamylase synthesi...
متن کاملThree isoforms of isoamylase contribute different catalytic properties for the debranching of potato glucans.
Isoamylases are debranching enzymes that hydrolyze alpha-1,6 linkages in alpha-1,4/alpha-1,6-linked glucan polymers. In plants, they have been shown to be required for the normal synthesis of amylopectin, although the precise manner in which they influence starch synthesis is still debated. cDNA clones encoding three distinct isoamylase isoforms (Stisa1, Stisa2, and Stisa3) have been identified...
